Chemical synthesis on solid-supports revolutionised molecular biology by making rapidly available custom made bio-oligomers in high-purity in days. A similar step-change in high-throughput synthesis of drug-like molecules is needed to overcome a bottleneck in drug development. Established solid-supports offer either high loading (<6.5 mmol/g in hydrophobic solvents) OR wide-solvent compatibility (organic/water at <<1 mmol/g). The paper reports a poly(vinyl ether) resin support with unprecedented solvent compatibility from water to hydrophobic solvents and <8.5 mmol/g loading. This solvent compatibility is obtained whilst still achieving 20-fold loading levels over competitive systems (e.g. Tentagel resins).
