This article was cited as a ‘Must Read’ by F1000. It was the first to measure both control and response processes of mitochondrial respiration and ATP production in skeletal muscles during the transition from rest to exercise. It combined arterio-venous O2 concentration difference measurements with rapid serial biopsies during stimulated contractions in animal muscle, to show that a simple ADP-feedback system was not sufficient to explain the complex dynamics of muscle oxygen consumption during exercise: rather, a complex allosteric activation system was required. In recognition of the significance of this finding, the paper was supported by a letter in the journal.
