A New Dynamic in Mass Spectral Imaging of Single Biological Cells
Although the introduction of C60+ primary beams has enabled ToF-SIMS to provide a molecular depth profiling and 3D imaging capability, high lateral resolution performance, is severely limited by the pulsed nature of current instruments. A completely new instrument, J105, using a dc primary ion beam that overcomes many of the drawbacks is described. The stream of secondary ions is sampled using a buncher-harmonic-ToF arrangement. The concept is validated by studies of biological samples generating 2D and 3D images showing subcellular molecular localization while maintaining high mass resolution. Two £1m instruments have been installed in the US (2011) and Sweden (2012).
