This is an original work that presents 'proof of principle' in results. Its significance is that the widefield surface plasmon resonance (WSPR) microscope used here enabled interfacial binding images with significantly higher lateral resolutions than prism based systems. We demonstrated the functionality of the WSPR by imaging a sequence of binding events between micro-patterned ECM proteins and their specific antibodies. Using the WSPR system a change in contrast was observed with each binding event. This work has provided better understanding of protein-surface interactions at the interface.
