Aberration-free three-dimensional multiphoton imaging of neuronal activity at kHz rates.
A fundamental problem in microscopy is that microscopes only image well features lying in the focal plane. We have developed a technique that allows images to be obtained along arbitrary three-dimensional trajectories. The master patent on this (US8498048) has been licensed to Carl Zeiss (www.zeiss.com, details available). This paper describes a neuroscience application where a focussed beam of light is scanned at high speed, without aberration, along an axon. Fluorescence is used to study an action potential travelling along the axon. This led to an MRC grant (MR/K015877/1 £1.2M –jointly with the Department of Physiology, Anatomy and Genetics, Oxford).
