Scale-up of protein purifications using aqueous two-phase systems: Comparing multilayer toroidal coil chromatography with centrifugal partition chromatography
The paper is the outcome of a follow-on grant (BB/FOF/206) from the BBSRC protein purification study (BB/C506364/1). A new multilayer toroidal coil centrifuge (MTCC) was developed to overcome the loss of stationary phase found when using a centrifugal partition chromatography (CPC). It was found that stage efficiency rose dramatically (up to 25%) as the centrifugal force field was increased. Furthermore there was no loss of stationary phase, with the MTCC having five times the throughput of the CPC with the same resolving power.
