Mechanistic modelling can be used to understand the fundamental behaviour of biological species during their chromatographic separation and purification. This paper describes the first example of modelling these separations at bead level. The model takes into account the heterogeneity in bead sizes and the spatial variations in the packed bed, thus offering a detailed description of the flow field and mass transfer phenomena. It provides the basis for in silico experiments to evaluate the design of new resins prior to lengthy experimental programmes to fabricate and test such materials. The work underpins ongoing studies with Pfizer (Dr Ranga Godavarti, Ranga.Godavarti@pfizer.com).
