An efficient approach to bioconversion kinetic model generation based on automated microscale experimentation integrated with model driven experimental design
Detailed kinetic models underpin successful bioconversion process design and optimisation but can be difficult to establish due to complex enzyme mechanisms and substrate/product inhibition effects. This work established a numerical approach for quantification of 6 independent kinetic parameters from a minimum number of experiments. When combined with automated microscale experiments this led to a 6-fold decrease in the number of experiments required (compared to conventional methods) and a 45-fold decrease in material requirements. This work formed the basis of our IChemE Innovation and Excellence award in Bioprocessing in 2010 and helped underpin the BiCE impact case study.
