Characterization of lentiviral vector production using microwell suspension cultures of HEK293T-derived producer cells.
This work examined the production of lentiviruses as a therapeutic vaccine for the treatment of Parkinson’s disease. A key challenge is to ensure that the virus suitably 'packages' the corrective gene and is biologically active for delivery. This paper combined statistical Design of Experiments with micro-scale cell cultures to identify key parameters for the production of lentivirus vectors. An optimal harvest window was identified based on an interaction between fluid hydrodynamics and virus half-life calculations. These findings currently underpin the development of a manufacturing-scale process for Phase 3 clinical trials by Oxford Biomedica (Dr Kiri Mitrophanous, k.mitrophanous@oxfordbiomedica.co.uk).
