This paper shows how changes in the shape of the micro-algae Euglena Gracillis were quantified with image processing techniques and the results were ranked against human (manual) cytometry. The methods that coincided with human observation were eccentricity measurements, cross-correlation without high-pass filtering and cross-correlation of the power spectral density. A unique, simple method to identify cell death was discovered by analysing the intensity standard deviation. The work is of particular relevance as the organism offers an abundant source of biofuel, and consequently resulted in an invited presentation at the International Conference on Biodefense and Natural Disasters (Orlanda, USA 2013, www.omicsgroup.com/conferences/biodefense-natural-disasters-2013/).
