Efficient Site-Specific Radiolabeling of a Modified C2A Domain of Synaptotagmin I with [Tc-99m(CO)(3)](+): A New Radiopharmaceutical for Imaging Cell Death
Protein based molecular imaging agents are usually labelled non-site-specifically; rarely is it demonstrated that labelled proteins retain full bioactivity; indeed the most damaged molecules carry the highest radiolabel signal. This paper describes an improved amino acid tag which, when part of any recombinant protein, leads to fast, efficient and site-specific labelling without further modification of the protein. This technology lends itself for high throughput screening of libraries of diagnostic and therapeutic proteins in vitro and in vivo, and we demonstrated this by developing a new radiopharmaceutical for imaging cell death. It was patented and further optimisation led to second patent.
