Precipitation is an important operation in biopharmaceutical purification yet the mechanism of protein precipitation in multi-component solutions is not well understood. Existing models lack fundamental understanding of the process. Here a new model is developed based on how the protein solubility changes during the process. The model structure is generic and robust and adequately reflects the non-linearity of protein precipitation kinetics. It thus provides new fundamental insights to protein precipitation in multi-component, complex protein solutions and can be applied to industrial processes to improve process design, model-based optimisation and process control.
