Prediction and verification of centrifugal dewatering of P. pastoris fermentation cultures using an ultra scale-down approach.
Yeast cells like Pichia are favoured in the bioprocessing industry for their ability to produce and release soluble therapeutic proteins of high quality. A challenge with Pichia are the very high cell densities achieved. This creates a bottleneck in the dewatering of the cells during separation where operating conditions at large scale need to be evaluated and optimised. This paper provides the first ultra scale-down predictive laboratory tool to assess the dewatering characteristics of cells during centrifugation. The work was sponsored by an IMRC partner, Merck, and verified at pilot scale in collaboration with Fujifilm Biotechnologies (Dr John Liddell, john.liddell@fujifilmdb.com).
