The STED method is revolutionizing optical microscopy by providing resolutions in the 10s of nanometers range, far smaller than the physical diffraction limit of light. However, full 3D resolution is very difficult to obtain, due to optical aberrations from specimens. This paper showed the first demonstration of specimen aberration correction, enabling 3D STED imaging deep in biological tissue. These results have already led to over ten invitations to speak at conferences, including a plenary talk at Focus on Microscopy 2013.
