A mathematical model of brain tumour response to radiotherapy and chemotherapy considering radiobiological aspects
Temozolomide (TMZ) has produced the largest single improvement in the outcome for brain cancer in 50 years, although the mechanism is not well understood. This paper shows that the effect of TMZ chemotherapy can be included in our brain cancer model. We show that TMZ appears to act as a radio-sensitizer and is not an independently cytotoxic agent. We also separate the effects of single and double strand DNA breaks in the model and show that the main effect of TMZ is on the repair of single strand breaks. This paper will accelerate the widespread use of combined TMZ and radiotherapy.
