We performed for the first time thermo-convective PCR with asymmetric primers leading to single-stranded products, We combined this with electrochemical detection after labelling the products with osmium tetroxide bipyridine. Thermo-convective PCR takes place near a heated platinum or gold microwire. Any PCR machine is not needed, rather, each volume element of the 50 microliter PCR solution experiences thermal cycling by mere thermo-convective movement through regions of different temperatures. This paves the way for extremely simplified and, yet, sensitive quantitative genosensors.
