Modelling in vivo skeletal muscle ageing in vitro using three-dimensional bioengineered constructs
Degeneration of skeletal muscle with age is a major contributor to functional decline, morbidity and mortality. Using an in vitro 3D bioengineered constructs with multiple population-doubled(MPD) murine myoblast we have shown that MPD had reduced myotube size, decreased contractile ability, response to force, reduced remodeling , decrease in functional genes and impaired regenerative potential required. Similar to ageing/atrophied muscle in vivo, thus providing a test bed for therapeutic interventions to prevent muscle degeneration with age. This is the first reported 3D model of an ageing muscle. The author made a substantial contribution to the conception and design of the study.
