A 1-step microplate method for assessing the substrate range of l-α-amino acid aminotransferase.
The amino group is important in pharmacological compounds because it gives biological activity. Aminotransferases are enzymes that can be used to introduce amino groups into pharmaceutical compounds; there are many thousands of them but the substrate range cannot be inferred from the protein sequence and it remains an early bottleneck when selecting an enzyme for industrial application. We developed a rapid first round characterisation method applicable to the broad range of aminotransferases which can be carried out in high throughput mode in microplates. This approach helped underpin many of the outputs of the BICE impact case study.
