This paper presents a novel experimental methodology employing cell nuclei as micro-markers, in conjunction with a rigorous modelling approach, facilitating a 3D reconstruction of the strain field within the microstructure of aortic valves for the first time. The potential application of the devised methodology to map strain fields in valvular tissues under biaxial loads, and adoption for other connective tissues such as tendons, has since been explored and developed at Queen Mary and UoP.
