<28> In Next Generation Sequencing technology it is important to distinguish putative natural variants (SNPs) from sequencing- or other errors. This study shows that the detection of SNPs (“SNP calling”) is affected by the identity of surrounding nucleotides. Around 80% of false positive SNPs can be corrected with the “second best call" probability, conditional on that of the first call. The study was carried out in collaboration with the Wellcome Trust Sanger Institute in Hinxton (Cambridge), a world leader in the field of DNA sequencing.
