A Multidisciplinary Approach Toward the Rapid and Preparative-Scale Biocatalytic Synthesis of Chiral Amino Alcohols: A Concise Transketolase-/omega-Transaminase-Mediated Synthesis of (2S,3S)-2-Aminopentane-1,3-diol
This work was the first to describe the integration of enzyme engineering, bioinformatics-led strategies, automated microscale bioprocessing, modelling and scale-up expertise with the aim to develop a concise and efficient two-step biocatalytic preparative synthesis of a chiral pharmaceutical intermediate from inexpensive substrates. The use of automated microscale technology facilitated rapid optimisation at the early stages of process development, when intermediates and information on reaction kinetics is often scarce, allowing successful process design and scale-up. The research has led to our IChemE Innovation and Excellence award in Bioprocessing in 2010 and helped underpin the BiCE impact case study.
